Description |
A ready to use PCR master mix, without primers and template. Each 25µl of master mix will give a 50µl PCR containing 1.25u SuperTherm Gold DNA polymerase mix, 200µM dNTPs, MgCl2, 1 X buffer. |
Stability |
The master mix is stable when stored at -20℃ for 12 months in a constant temperature freezer or at 4℃ for up to 3 months. The master mix will survive a minimum of 10 freeze/thaw cycles. Shipped on ice. |
Supplied Readents |
2X Mster Mix, supplied as a 10ml aliquot sufficient for 400 rections.
1 X 25mM MgCl2 |
Use |
Thaw vial, mix and place on ice. Remove 25µl and add primers, template and extra MgCl2 (if required) in volume of 25µl to bring the total reaction volume to 50µl, Long term storage of the master mix should be at -20℃, not at room temperature. If reactions are not to be used immediately, keep them on ice for a maximum of 3 hours. |

Aplications |
This enzyme is designed for use in hot start PCR applications. The enzyme is supplied in an inactive state and requires activating. This is achieved by the addition of a single 10min, 95℃ pre-incubation step at the beginning of the end-users thermal cycler programme. |

Unit Definition |
One unit of enzyme is defined as the amount of enzyme that will incorporate 10nmoles of dNTP’s into acid insoluble material in 30 minutes at 70℃ under the assay conditions below. |
Unit Assay Conditions |
25mM taps (pH 9.3 at 25℃), 50mM KCl, 2mM MgCl2, 1mM B-mercaptoethanol, 200µM each dATP, dCTP (a mix of unlabelled and (3H) dCTP), dGTP, dTTP, 12.5µg activated calf thymus DNA, in a total volume of 50µg. |

Quality Control |
Each lot is tested for activity in PCR. Using lambda DNA as template we guarantee an amplification of at least 10 |
